چکیده

The sipunculid, Themiste pyroides is found intertidally to subtidally from Southern British Colombia to Northern Mexico. An avid burrower in rock nooks and crevices, this worm is prone to hypoxic environments, particularly during low tide. T. pyroides uses the oxygen-transport protein, hemerythrin to transport oxygen from the tentacles to the vascular system and into the coelom. Because hemerythrin changes to colorless when deoxygenated, the degree of saturation could be measured using spectrophotometry. This study looks at three replicates of purified T. pyroides hemerythrocytes and the degree of saturation with pH, to assess the possibility of a Bohr shift. Conclusively, no Bohr shift can be identified at this time due to the variation in the number of cells. A study looking at absorbency of multiple hemerythrocyte concentrations would serve worthwhile to eliminate this variation. Also, future studies could be done on assessing the presence of a Root effect.

Amino acid sequence of hemerythrin from Themiste dyscritum.

The amino acid sequence of hemerythrin from the sipunculid worm, Themiste dyscritum, was determined by sequenator analyses of the S-pyridylethylated protein and fragments derived by further chemical and enzymatic cleavages. The fragments were obtained by cleavage of the intact protein with hydroxylamine, trypsin digestion of citraconylated intact protein, and subdigestion with Staphylococcal pr...

Molecular Phylogeny and Surface Morphology of Marine Archigregarines

The trophozoites of two novel archigregarines, Selenidium pisinnus n. sp. and Filipodium phascolosomae n. sp., were described from the sipunculid Phascolosoma agassizii. The trophozoites of S. pisinnus n. sp. were relatively small (64–100mm long and 9–25mm wide), had rounded ends, and had about 21 epicytic folds per side. The trophozoites of F. phascolosomae n. sp. were highly irregular in shap...

The reduction of methemerythrin by e-aq and CO2- from pulse radiolysis studies.

The rate constants for reduction of methemerythrin from Phascolopsis gouldii and Themiste pyroides by hydrated electrons are 2.0 and 3.9 x 10(9) M(-1)s(-1), respectively, at pH 8.2, I = 0.03 M, and 25 degrees C. There is only a small increase in rate when the pH is lowered to 6.3 and a very small decrease when the ionic strength is raised to 0.1 M. Adding azide ion (to form the met-azide adduct...

Influence of solvent accessibility and intermolecular contacts on atomic mobilities in hemerythrins.

Thermal factor parameters (B values) have been compared from the refined crystal structures of the myohemerythrin from Themiste zostericola and of the octameric hemerythrin from Themiste dyscrita. These B values, which are directly related to atomic mobilities, were found to correlate rather closely with the solvent accessible areas within the respective crystals. Although protomeric units of t...

Clot Formation in the Sipunculid Worm Themiste petricola: A Haemostatic and Immune Cellular Response

Clot formation in the sipunculid Themiste petricola, a coelomate nonsegmented marine worm without a circulatory system, is a cellular response that creates a haemostatic mass upon activation with sea water. The mass with sealing properties is brought about by homotypic aggregation of granular leukocytes present in the coelomic fluid that undergo a rapid process of fusion and cell death forming ...
